A polyketide synthase PKS was generated by genomic recombination of a section of a macrolide producing PKS from Penicillium patulum followed by 3 domains from an aromatic producing PKS isolated from Fusarium graminearum This recombinant PKS could be expressed in E coli and a series of feeding experiments were undertaken When the E coli were cultured with a source of acetate but starved of methionine the polyketides 4 5 and 6 were isolated When the polyketide 4 was added to fresh E coli culture that was starved of acetate but fed methionine then a series of polyketides containing either 19 20 or 21 carbons was obtained When the E coli were cultured with sources of both acetate and methionine polyketides were produced with even higher numbers of carbons incorporated OH O HO OH O OH HO 5 OH HO OH O HO 6 O OH OH derive the structure of the recombinant polyketide synthase PKS which produces compound 4 The answer would include the following 1 the total number of domains ii the sequence of enzymes that would be found in each domain iii the structure of the polyketide chain that is attached at the start of each domain iv the structure of the final linear polyketide
